The techniques vary in detail but not in concept. Typing sera for histocompatibility antigens are not yet as clearly defined as are those for typing red cells. Histocompatibilityantibodies develop in the sera ofpatients who haverej ected tissue grafts, of patients who have had multiple blood transfusions, and of women who have had multiple pregnancies and have been immunized by their offspring. These sera are used to type lymphocytes ofpotential organ graft donors and recipients. If the lymphocytes of both donor and recipient are agglutinated or killed by a given typing serum, then as far as that serum is concerned thelymphocytes of both individuals have antigens in common.
Undoubtedly some of these cases are due to inadequate serological techniques, but another possibility is that serological typing gives no information on the immune potential of the recipient, which can be an important variable. Lymphocyte culture techniques may help in this assessment and they will now be discussed. LYMPHOCYTE CULTURE REACTIONS If blood lymphocytes from two individuals are cultured together in a nourishing medium they grow and divide. The proliferative activity can be assessed by measuring the incorporation of radioactive labelled materials by the cultured cells.
